Indian skipper and opener Rohit Sharma smashed 40 runs off 24 balls while another opener Shubman Gill scored 23 runs from 24 balls, Suryakumar Yadav smashed 22 runs off 14 balls and Ravindra Jadeja smashed 29 runs off 15 balls.
South African bowlers Lungi Ngidi, Marco Jansen, Kagiso Rabada, Tabraiz Shamsi and Keshav Maharaj got one wicket each.
Earlier, India won the toss and opted to bat first.
India is the topper in the point table of the tournament with 14 points followed by South Africa with 12 points.
